Acquisition of Equipment and Infrastructure at ISABIAL: Leica Thunder 3D Assay Microscope

Project reference number: IFEQ22/00118
Brief description of the objective or purpose of the grant:

Acquisition of Scientific Infrastructure. Leica Thunder 3D Assay Microscope comprising: (1) Fully automated microscope with transmitted light in brightfield and phase contrast, and epifluorescence, incorporating computational clearing technology. (2) LED8 illumination with an emission filter wheel for 8 LED excitation bands and 8 emission bands (from UV to IR) and transmitted light. (3) Motorised fluorescence turret for red, green, blue and far‑red channels. (4) Motorised stage for high‑speed XY scanning with inserts for slides, Petri dishes and multi‑well plates. (5) Motorised focus with closed‑loop control and Z‑focus capability (AFC autofocus control system). (6) Quantum HS stage combining high‑speed movement with maximum image quality through an optimised motion algorithm ensuring smooth, vibration‑free operation. (7) Objectives: 5x, 10x, 20x air, and 40x and 63x oil, with phase contrast. (8) Thunder 3D DCV Tera module to facilitate 3D image acquisition and eliminate blurred or out‑of‑focus images. (9) Leica K5 high‑sensitivity camera. (10) Computer with 32″ monitor and AIVIA GO software for image acquisition and subsequent analysis. (11) On‑stage incubation system and incubation chamber for cell plates, with splash‑protection system enabling live‑cell imaging.
The equipment will be installed in the multipurpose room of the Research Support Laboratory (LAI), and later transferred to the imaging unit of the new ISABIAL building, adjacent to the new Cell Culture Platform.
